| Abstract: | The present invention relates to attenuated salmonella and its use in treating tumors. The present invention belongs to biological technique field. The present invention discloses the constructions of pH1Si-Stat3, pGRIM-19, and pGRIM-19-Si-Stat3 recombinant plasmid, which coexpresses GRIM-19 and siRNA-Stat3. An attenuation salmonella electrotransformated with the recombinant plasmid is disclosed. The coexpression of GRIM-19 and siRNA-Stat3 has a good synergistic inhibition effect on the growth and metastases of tumors in the experiment in vivo. Attenuated salmonella has phagorytosis effect on tumors, and the progenitive specificity in tumortissue is more than 1000 times as that in normal tissue. The attenuated salmonella carrying effective plasmid, which can coexpresses siRNA-Stat3 and GRIM-19, can reach the deep part of tumors, and thus markedly inhibit their growth and metastases and evidently prolong the survival time of mice. | |||||||||||
